Note: Due to exceptional performance issues, this puzzle has been made worth zero points. Players should not feel compelled to play this puzzle to remain competitive on the leaderboards. However, if you are able to play this puzzle without issue, we'll still analyze any solutions.
This symmetric design puzzle has C2 symmetry, with two symmetric chains. This puzzle is a little different from other symmetry puzzles, in that we want players to focus on building a smaller interface between the two chains. There is no H-bond Network Objective in this puzzle, so the interface can be completely nonpolar (orange sidechains). We want to see if Foldit players can still build strong interfaces while limiting the number of nonpolar residues on the surface of each chain. See the puzzle comments for Objective details. The Baker Lab will run folding predictions on your solutions for this puzzle, and those that perform well will be synthesized in the lab. Remember, you can use the Upload for Scientists button for up to 5 designs that you want us to look at, even if they are not the best-scoring solutions!
Core Existence: Monomer (max +2100 +2260)
Ensures that at least 21 residues are buried in the core of the monomer unit.
Core Existence: Complex (max +2100 +2260)
Ensures that 42-52 residues are buried in the core of the entire complex (including the interface between monomer units). Players will not receive the full bonus if more than 52 residues are found in the core of the complex.
SS Design (max +500)
Penalizes all CYS residues. Penalizes GLY, ALA residues in sheets and helices
The game is freezing for 0.5-1.5 seconds whenever I drag in a building block, move around a cut segment, Freeze a segment, or other related matters. I'm assuming it's because the game has to do double duty for rendering and calculating the dimer ghost's score, but I wish this puzzle had a mode where you can edit a monomer, then introduce the dimer.
Edit: I've realized after the fact that this is in part due to having filters on at all times (I typically can handle performance impacts from filters but this is one of the exceptions), but even with all filters toggled off there is a 0.1-0.3 second freeze between actions.
We're sorry to hear the symmetry puzzles are still having some performance issues! This has been a tough problem to solve, but your continued feedback is helpful!
Also, thanks for noting the error in the maximum Objective bonus; I've corrected the comment above.
