Description

We are giving you a single chain of 76 residues to design. Nearing the end of this puzzle, we will post a 76 residue symmetric dimer puzzle where you will be able to load your solution from this monomer into the dimer. Keep that in mind when you are designing your monomer.

There are four filters enabled on this puzzle. The ResidueIE filter ensures PHY, TYR, and TRP residues are scoring well. The 1st LayerScore filter ensures the number of residues in the core is between 30% and 70%. The 2nd LayerScore filter ensures the number of residues on the surface is between 30% and 70%. The 3rd LayerScore filter ensures the number of boundary (residues between the core and surface) residues is below 70%. We hope these filters will help you design proteins that the scientists will be able to create in the lab.

The Layer Score filter ensures the proteins will fold up into desirable and realistic shapes. It will categorize each backbone residue as either 'core', 'surface', or 'boundary'. If you think of the protein as being suspended in a liquid, the 'surface' residues are those most exposed to the liquid. Subsequently, the 'core' residues are those that are most protected and the 'boundary' residues are the ones that lie between the 'core' and the 'surface'. Most residues are categorized as either 'core' or 'surface'.
