Description

Our 900th Foldit puzzle might bring up some Déjà vu. Not only will this 62 residue amino acid sequence look familar, but so should the starting model. This model was the best server prediction for target T0759, generated by the nns server. We've been told that ~45% of this model matches the native! More exciting info in the puzzle comments.

If you recall, we had to close and repost the first CASP WeFold puzzle to add in a 6th server model. That particular model was a very different topology compared to the other 5 server models.

Well, it's a good thing our WeFold collaborators noticed that extra model, because it is that exact server model that the CASP organizers selected for refinement target TR759!

The only difference is that they have trimmed both ends of the protein: http://predictioncenter.org/casp11/target.cgi?id=43
"Starting model's GDT_HA=45 (calculated on res. 46-107). Residues 1-12 and 108-109 are absent from the experimental structure and deleted from the model. Additionally, we deleted residues 13-45 as they do not form contacts with the rest of the structure."

Because many of you already used model 6 in 890b: CASP11 T0759: WeFold Round, we are making this refinement puzzle a bit different by enabling High Power Wiggle.

As we explained in the recent Wiggle Power Blogpost, we decided to give High Wiggle Power a rest during CASP11. However, the results we showed in that blogpost were based on Predicted Contacts & De-novo puzzles, so this will be a good test for how useful High Power Wiggle is for Refinement.

Hopefully the native for target 759 will be released shortly after its CASP deadline, so we will be able to see whether or not we should continue to use High Power Wiggle for future CASP11 refinement puzzles.
