On 2232 for example, manual mutate is disabled, and I now I have 3 cysteines and a disulfide bond in my design. But I can't change it so I'm penalized 150 points. Seems to me that if you are allowing J or Y to use cysteines with no way to fix it, there should be no penalty. Or just enable manual mutating M.
Sorry I didn't see this before Puzzle 2232 closed! Hmm, manual mutate should not be disabled on Puzzle 2232 (nor any other protein design puzzle). Can you double check that manual mutate is truly disabled?
A little while back we updated the manual mutate icon to a yellow trace like this:

If you're used to using the keyboard shortcuts, it's possible that the shortcut in your client was changed to something other than M. You can verify or edit shortcuts in the Foldit options under General Options -> Controls -> Configure Keyboard Shortcuts. For manual mutate, scroll down to "Actions: Mutate the segment".

HI @bkoep I don't use any exotic shortcuts ever, but, I checked your path and I the line reads: "Actions: Mutate the segment M " I have a snip which I will show you in discord since I can't figure out how to attach it or paste it in here. It shows that when I select M, the message reads " mutate the segment " but as you can see the entire panel of AAs is not live for picking. I will also try to send the expired 2232 to scientists in case you can look at it.
Thanks for following up @alcor29, that's definitely a bug! It seems to be specific to disulfides and easy to reproduce, so hopefully it should be easy to track down and fix.
In the meantime, one workaround is to break the disulfide first (for example, use Pick Sidechains to move one of the CYS sidechains away from the other). Then, you may be able to mutate the CYS residues to something else.
