In Foldit small molecule design puzzles, if we made a ligand with -SH groups on it (like in cysteine), could the ligand form disulfide bonds with nearby cysteine sidechains from the protein?
In Foldit small molecule design puzzles, if we made a ligand with -SH groups on it (like in cysteine), could the ligand form disulfide bonds with nearby cysteine sidechains from the protein?
We don't annotate ligand -SH groups as "disulfide-able", so Foldit won't ever make the protein-ligand disulfide connection.
(That's not to say such a bond couldn't happen with an actual drug - it's just that we don't really consider covalent connections between ligands and proteins currently within Foldit.)
