Description

This puzzle challenges players to design proteins with more sheets! Designs should include at least 50% residues in sheet conformation. The Ideal Loops Filter is meant to encourage the use of the Ideal Loops that are displayed in the Rama Map. See the puzzle comments for filter details. The Baker Lab will run folding predictions on your solutions for this puzzle, and those that perform well will be synthesized in the lab. Remember, you can use the Upload for Scientists button for up to 5 designs that you want us to look at, even if they are not the best-scoring solutions!

Residue IE Score: Monitors that all PHE, TYR, and TRP residues are scoring well.

Core Existence: Ensures that at least 30% of the residues are buried in the core of your design.

Secondary Structure: Checks that at least 50% of residues are in sheets; penalties are incurred if fewer than 50% of residues form sheets.

Secondary Structure Design: Penalizes all CYS residues. Penalizes GLY, ALA, and PRO residues in helices and sheets.

Residue Count: Penalizes extra residues inserted beyond the 75 residue limit. Players may insert up to 10 extra residues.

Ideal Loops: Penalizes loops that do not conform to preferred ABEGO patterns. Use Auto Structures to see which residues count as "loops."

this puzzle makes no sense to me, tried a few different combinations and most of them are not getting past the sheet filter

I'm also finding it hard to get past the sheet filter. However two designs shared in my group have passed the filters. One is nearly symmetrical and the other (better scoring design) is not.

I realize you guys have real world things coming up but your alert that you'll be shorthanded comes at a VERY bad time… there are serious game issues that haven't been resolved to the point that puzzles that should be fine are not. 1265 keeps crashing on me, the IL and Sheets filters are making this puzzle much harder (and perhaps more crashable) than it should be… and you're getting ready to leave us with little help… this doesn't make sense to me and is very discouraging. I have played for about 18 months now and have not yet experienced the difficulties (in simply playing, not in challenges to my skills) that I have recently experienced, and I am rethinking Foldit as a way to spend my spare time because of it. HELP!

… just that li'l correction to the above. Might not be the right place to post this, but maybe so as this particular puzzle is crashing most frequently for me.
