BIOTIN!! Now with 60% more fun! This time, the scaffold you'll design in is a rat intestinal fatty acid binding protein (PDB: 1icm). There is one constraint in place to encourage the biotin tail to face towards the solvent (because in most applications, biotin has a linker connected to that tail that needs to get out). You're allowed to move the backbone a little bit , but don't overuse that backbone wiggle or else you'll have a hard time getting it back. (Use sidechain wiggle instead) SCORING: The ligand score is the primary contributor to the puzzle score. However, the other designable residues also contribute to a much less extend. Finally, the designable residues scores are also weighted based on their occurance in the multiple sequence alignment of the original scaffold.
