Description

We are giving you 3 linked residues and your job is try to design and fit them into a constrained region of the CTLA-4 protein. We know the general region in which we want you to place to residues, but we don't know exactly where is the best spot; look for good hydrophobic pockets or residues to make strong hydrogen bonds to. If you are new to Foldit, make sure you have completed Intro Puzzle 5-2, and 7-1 through 7-4 before trying this puzzle.

It is not a start point, I`m able to load my gallery solution…
Dev client automagically loaded my best state on start.
As far I remember it should be impossible?

The great news is that already (after just 1 day) your Foldit solutions on this puzzle look really promising, we are very excited!!!

The results so far look very interesting, some of them are quite elegant! As a lot of you already discovered the pronounced cavity is a nice keyhole to use :). I hope the Easter weekend can give you even more inspiration. Keep up the good work!

I have a general question which fit quite good in this puzzle. How do you chose the solutions you gonna work with? Simply best scoring? Diversity?
Diversity is a big thing in this puzzle, huh?
I have now a solution made offline which seem different, than my other solution AND it is still good scoring. Is it sufficient to work on it a bit more so that it is saved on the server. Do you save all the solutions generated or just the best scoring ones?

This translates to about 500,000 Foldit solutions per puzzle!

As you can imagine that is way too much to look at so we do filter a lot. The different filters depend on the puzzle type, but here are some general ones:

1) Examine the top-scoring soloist solutions for every player
2) Examine the top-scoring evolver solutions for every player
3) For design puzzles: Examine the top-scoring solutions for each unique sequence
4) Cluster the solutions and look at the most populated clusters (this can sometimes be a problem if one team produced many similar solutions) so sometimes we have to cluster in a different way:
5) Take the top-scoring solution and look for the next highest-scoring solution that has an RMSD of at least X Angstroms, and cluster that way down the list. This gives us many different high-scoring topologies to look at.

So for this particular puzzle, if you have two very different solutions that score well, we will examine both of them for sure (as long as they are structurally different from one another or have a different sequence).
