Description

This is a 60 residue symmetric tetramer design puzzle; there will be four symmetric chains to manage. This particular type of symmetry is a ring (C4) symmetry. Remember, a symmetric protein relies not only on how well-folded each chain is, but also on how well they interface together. See the blogpost about this: http://fold.it/portal/node/993871

There are 4 filters enabled in this puzzle. They are slow to compute, so you may want to check "Disable slow filters" in the behavior tab for when you are manually pulling and running your scripts. You should remember to turn the filters back on though, because only then will you see your true score.

The ResidueIE filter ensures PHY, TYR, and TRP residues are scoring well. The "Core Exists" filter ensures the number of residues in the core is between 30% and 70%. The "Polar Core" filter that the residues in the core are properly hydrophobic. The "Layer Design" filter looks at the interactions of the residues based on the surface area of the protein. Residues can be either in the core, on the boundary or on the surface of the protein. Depending on the surroundings, the "Layer Design" filter will give you a bonus if your amino acid matches what it was expecting based on an analysis of the surface area of the protein.

Due to the new filters slowing things down, and since there are many design puzzles up, we've extended this deadline by 1 week.

FYI: I ran mutate for 8 hours with filters on, and at the end, I have "Too many polar residues in the core" and 19 invalid residues. (monomer is all helices).

Just want to report to the devs that once again, even with the filters, a megahelix scores far higher than any realistic structure.
