Similar to the previous puzzle in this series, this protein has had its core changed to Alanine for you to mutate. The loops are still locked, but this time there is no inserting allowed! Constraints are holding the backbone near where it starts out, so focus on the sidechains.

The native for Mini-CASP 6 was released and we will post the Quest to the Native soon. We noticed that a few players got very close to the native, but were not able to get a high Foldit score. We are giving you 4 different solutions from Quick Mini-CASP 6 Puzzle to see if you can improve the Foldit scores. Each time you reset the puzzle one of these 4 Foldit predictions will be randomly chosen.

The solved structure for Mini-CASP 3 will be released soon. We hope you can improve on this Rosetta prediction.

BIOTIN!! Now with 60% more fun! This time, the scaffold you'll design in is a rat intestinal fatty acid binding protein (PDB: 1icm). There is one constraint in place to encourage the biotin tail to face towards the solvent (because in most applications, biotin has a linker connected to that tail that needs to get out). You're allowed to move the backbone a little bit , but don't overuse that backbone wiggle or else you'll have a hard time getting it back. (Use sidechain wiggle instead) SCORING: The ligand score is the primary contributor to the puzzle score. However, the other designable residues also contribute to a much less extend. Finally, the designable residues scores are also weighted based on their occurance in the multiple sequence alignment of the original scaffold.
