Ever get a hankering for redesigning a glycerolphosphate synthase into a biotin binding protein? I sometimes do, which is why I've made this nice puzzle for you. This puzzle also has a single constraint meant to encourage the biotin tail to face outwards into the solvent. (Imagine a linker is attached at the tail end). You're allowed to move the backbone a little bit in this one too. SCORING: The ligand score is the primary contributor to the puzzle score. However, the other designable residues also contribute to a much less extend. Finally, the designable residues scores are also weighted based on their occurance in the multiple sequence alignment of the original scaffold.

It's your favorite B vitamin! Biotin! This time it's trying to find a home in a tricorn-interacting factor F1 protein. (PDB: 1xrl) You can still move the backbone a little bit, but be careful about wiggling the backbone too much. SCORING: The ligand score is the primary contributor to the puzzle score. The submission deadline for Mini-CASP6 got pushed back so we wanted to give you Rosetta@home's latest prediction to work on (in addition to puzzle 214's predictions). This puzzle will expire on Halloween unless the deadline gets pushed back even further.

This is the last Mini-CASP6 puzzle before we find out what the correct structure looks like. Since this is a difficult de-novo target, we wanted to give you some Foldit predictions (from the freestyle and multi-start puzzles) to see if you can improve on them at all. It might be hard to improve the Foldit scores on these 5 models (since these were top-scoring Foldit predictions) but hopefully seeing another player's solution might lead you to a winning prediction!
