This is the final mini-CASP puzzle before CASP9 starts on monday. Here are 6 new templates for mini-CASP 23. Since it is such a large protein we are giving you some extra time to use the alignment tool for each of the 6 templates.

Here is a target from CASP8 designed to train you for CASP9. We are giving you the 2 best possible templates, with 2 different alignments for each. The native is shown as a guide. We hope that this puzzle will be useful for you to see what alignment tool moves are needed to reach the native state. Note: this puzzle expires this friday!

Here's the next puzzle in the Hold it! series. This protein is binding a small molecule, and forming several hydrogen bonds at one end. This time things are already in a good position to form the hydrogen bonds, and they are held in place with constraints. We need you to mutate the residues around the small molecule to make better contacts! This puzzle will be relatively short!

This puzzle is the same as Mini-CASP 21, but with an insertion of 8 residues in the middle of the protein. We are giving you 7 different templates to work with on this puzzle (and 2 other templates in puzzle 294).
-
This puzzle is the same as puzzle 293 but these 2 templates are very different than the 7 templates in puzzle 293. To make the other 7 alignments easier to see in the alignment tool of the other puzzle, this puzzle only has these 2 complicated alignments.

Here is the third of 3 top scoring Rosetta@home predictions that you can start from. Drastic moves might be necessary to get closer to the native with this one, so make sure to look at starts from mini-CASP 19a and 19b for differences.

Mini-CASP 19 is a tough de-novo target. Here is the second of 3 top scoring Rosetta@home predictions that you can start from. Drastic moves might be necessary to get closer to the native with this one, so make sure to look at 19a and 19c for comparison.

Mini-CASP 19 is a tough de-novo target, where we have no templates or alignments to start from. Here is one of 3 top scoring Rosetta@home predictions that you can start with. Some drastic moves will probably be necessary to get closer to the native with this one, so explore as much as possible. Both termini have been trimmed compared to puzzle 274.

Mini-CASP 22 had only 1 template with no alignments to the first 23 residues. This could mean the N-terminus is disordered, so we have trimmed off the first 13 residues (see puzzle comments for more details). We have also included 2 very distant templates, you can see that their scores are quite bad and the starting alignments won't thread, but maybe they can help somehow.
