This is the thirteenth CASP10 Refinement Target. This model was the best server prediction for target T0705, generated by the AOBA-server. We've been told that ~65% of this model matches the native, so you'll need to significantly change the starting model. Luckily, this is a short target with only 96 residues. This is the twelfth CASP10 Refinement Target. This model was the best server prediction for target T0723, generated by the Matrix server. We've been told that ~85% of this model matches the native, so you just need to find which 15% to refine! More info in the puzzle comments.

As mentioned in Puzzle 600, we are giving you server predictions for the last non-refinement CASP10 target. Resetting the puzzle will cycle through 8 different server predictions (that are similar to one another) and they are available in the Alignment Tool so you can use partial threading between them. Please try out each start, as the topology that initially scores poorly may actually be the closest to the native and will eventually score better than everything else.

This is a small (50 residue) symmetric dimer design puzzle. We saw some very interesting structures in the smaller 20 and 30 residue freestyles. Using 50 residues puts this in the realm of a full-scale protein domain, and docking large secondary structure motifs becomes more viable. Remember, a symmetric protein relies not only on how well folded each part is, but also on how well they interface together.

This is the eleventh CASP10 Refinement Target. This model was the best server prediction for target T0712, generated by the MUFOLD-Server. We've been told that ~92% of this model matches the native! So (for once) you won't need to change the starting model much. In addition, the organizers have told us "Regions worth specific attention: 43-52, 79-92, 104-118." These are highlighted in Note Mode.

The fourteenth CASP10 target is another big one: 165 residues and none of the alignment programs were able to find good templates for this target. It might be the case that this protein has no homologs. We are giving you 4 different templates (in hope that one of them is correct) for now, and will provide you with some server predictions soon. This is a small (30 residue) symmetric trimer design puzzle; there will be three symmetric chains to play with. We saw some very interesting structures for the related 20 residue puzzle (Puzzle 553), but ten extra residues, and three-fold symmetry, should make this a very different challenge. Remember, a symmetric protein relies not only on how well folded each part is, but also on how well they interface together - and a symmetric trimer can be thought of as having three 2-fold interfaces. Stay tuned for higher-order symmetries, like tetrahedra and octahedra!
