Because of this, we are giving you the Rosetta model as a template using the Alignment Tool minus the regions the CASP assesors have told us are incorrect; start by threading it and fixing the cutpoints. This is a small (40 residue) symmetric dimer design puzzle. We saw some very interesting structures for the related 20 residue puzzle (#553). Adding twenty residues puts this in the realm of full-scale protein domain, and docking large secondary structure motifs becomes more viable. Remember, a symmetric protein relies not only on how well folded each part is, but also on how well they interface together. Stay tuned for three- and four-fold symmetry (trimers and tetramers) and polyhedra!

This is a small (20 residue) symmetric dimer design puzzle. We're interested to see what sort of structures you can come up with that respect the dimer symmetry. Remember, a symmetric protein relies not only on how well folded each part is, but also on how well they interface together!

The eleventh CASP10 target is not an easy one. It's 152 residues and none of the alignment programs were able to find good templates for this target. It might be the case that this protein has no homologs. We are giving you 6 templates (in hope that one of them is correct) for now, and will provide you with some server predictions once they are released. We are looking for the best locations to place & design FIVE linked residues on the surface of Influenza H3 Hemagglutinin. We have placed 2 helices on either side so that it will be easier to build scaffolds for your designs. Mutate the 5 sidechains while looking for good hydrophobic pockets or residues to make strong hydrogen bonds to. If you are new to Foldit, make sure you have completed Intro Puzzle 5-2, and 7-1 through 7-4 before trying this puzzle.
