For this 133 residue CASP11 Target, T0838, we are giving you 2 different server predictions selected by our WeFold collaborators. Resetting the puzzle will cycle between both aligned models and they are available in the Alignment Tool so you can use partial threading between them. For this target, the CASP11 organizers have provided additional "correct" contact predictions, which we have combined with the sparse data contacts from Puzzle 933b. Contacts are scored with the Contact Map filter in this puzzle, and there is an upper limit of 8850 possible points from the filter. Players may load in solutions from puzzle 933b. For this target, the CASP11 organizers have provided "sparse" experimental data about possible contacts in the protein. Contacts are scored with the Contact Map filter in this puzzle, and there is an upper limit of 6350 possible points from the filter. The starting structure is an extended chain, and secondary structure predictions can be found below in the puzzle comments.

This design puzzle introduces a new filter! The Secondary Structure filter counts the number of residues in helices or sheets by recognizing hydrogen-bonding patterns. Players will receive a 500 point bonus if fewer than 50% of residues are in helices; penalties are incurred if more than 50% of residues form helices. There are 3 additional filters enabled on this puzzle: the Residue IE Score monitors that all PHE, TYR, and TRP residues are scoring well; the Core Existence filter checks that at least 20% of the residues are buried in the core of your design; and the Fragment filter scans for parts of your design that are unlikely to fold up naturally. The Baker Lab will run folding predictions on your solutions for this puzzle, and those that perform well will be synthesized in the lab. Remember, you can use the Upload for Scientists button for up to 3 designs that you want us to look at, even if they are not the best-scoring solutions!

This CASP11 refinement target is an extremely large 251 residue TIM barrel. Beware that not all computers are powerful enough to run this puzzle comfortably. Due to player requests for some larger, more interesting CASP11 targets, we are making this puzzle available to all Foldit players who wish to work on it. However, players should not feel obligated to play this puzzle to maintain leader board rankings; this puzzle is worth only 10 points!
