This CASP refinement target has 143 residues, and we've been told that ~65% of this starting structure is correct. High Wiggle Power has been enabled for this puzzle.

***WARNING: This puzzle has been CLOSED & REPLACED by Puzzle 911b,***

NOTE: This puzzle replaces Puzzle 911, which included 2 additional starting models that our WeFold collaborators no longer trust.

This CASP refinement target has 76 residues. We've been told that ~60% of this starting structure is correct. High Wiggle Power has been enabled for this puzzle. Secondary structure predictions are in the puzzle comments.

This is a design puzzle with 70 residues. Similarly to Puzzle 898b, helices will not score as well as they normally do, and this time we've tweaked the Fragment filter to be less stringent. There are 3 filters enabled on this puzzle: the Residue IE Score monitors that all PHE, TYR, and TRP residues are scoring well; the Core Existence filter checks that at least 25% of the residues are buried in the core of your design; and the Fragment filter scans for parts of your design that are unlikely to fold up naturally. The Baker Lab will run folding predictions on your solutions for this puzzle, and those that perform well will be synthesized in the lab. Remember, you can use the Upload for Scientists button for up to 3 designs that you want us to look at, even if they are not the best-scoring solutions!

The CASP organizers have told us that this HIV-1 Env protein is a trimer, so we are posting a Bonus Round for T0787 as a symmetric trimer! We've provided you with a RosettaServer model to start with. More info in the puzzle comments about different possible folds.

For this target, the CASP11 organizers have provided "sparse" experimental data about possible contacts in the protein. Ironically, the "sparseness" of the data manifests as a huge number of possible contacts, with very little information about which contacts might be correct. Contacts are scored with the Contact Map filter in this puzzle, and there is an upper limit of 2029 possible points from the filter. Contact predictions are provided only for residues 31-160, so focus on that region of the protein! Players may load in solutions from Puzzles 888 and 894 and we've provided you with 5 Robetta server models. Restart the puzzle to cycle through the alternate starting structures.
-
NOTE: This puzzle replaces Puzzle 907, which included a much larger set of contact predictions. All contacts now have equal weight, and players should aim to satisfy as many contacts as possible.

This 153 residue CASP11 Target is part of the HIV-1 Env complex. We are giving you 4 different server predictions selected by our WeFold collaborators. Resetting the puzzle will cycle through each model and they are available in the Alignment Tool so you can use partial threading between them.
