The first CASP11 refinement target is larger than most Foldit puzzles, at 201 residues. This model was the best server prediction for target T0760, generated by the MULTICOM-NOVEL server. We've been told that ~58% of this model matches the native! As this is such a large protein, we are giving you 10 days to work on it. More info in the puzzle comments, where you can post your feedback about whether puzzles of this size are too much for us to tackle in CASP11.

This CASP11 target has only 70 residues! We've provided you with 5 Robetta server models. Resetting the puzzle will cycle through each model and they are available in the Alignment Tool so you can use partial threading between them. See the puzzle comments for secondary structure predictions!

This is a single-chain design puzzle with 80 residues, and eight of the residues are pre-folded into a beta-hairpin. These residues are somewhat flexible, but there are strong constraints to keep them close to beta-hairpin conformation. There are 3 filters enabled on this puzzle: the Residue IE Score monitors that all PHE, TYR, and TRP residues are scoring well; the Core Existence filter checks that at least 30% of the residues are buried in the core of your design; and the Fragment filter scans for parts of your design that are unlikely to fold up naturally. The Baker Lab will run folding predictions on your solutions for this puzzle, and those that perform well will be synthesized in the lab. Remember, you can use the Upload for Scientists button for up to 5 designs that you want us to look at, even if they are not the best-scoring solutions!
