This is the first of the many new exciting Flu puzzles David Baker mentioned in his blogpost. Our lab designed against the H1N1 subtype of the virus, but a related subtype with pandemic potential, H2N2, has different sidechains in this region that perturb binding. The goal is to design the interface of these designs so that it can bind to the H2 subtype of Influenza. This puzzle is worth 150 global points! If you are new to Foldit, make sure you have completed the Intro Puzzle 4-5 and Design Puzzles 6-1 through 6-4. We've tweaked some parameters since the last time you've seen this protein, so things should be a little different!

In electron density puzzles, we give you some experimental data to help you find the correct fold. This data takes the form of an electron density, and appears in game as a guide. You can adjust the guide visualization by going to [Actions]>[Electron Density].

We are giving you 5 server models as starts for this puzzle. Each time you reset the puzzle one of these 5 Zhang server predictions will randomly be chosen. They are also in the Alignment Tool so you can use partial threading and well as an extended chain. We would like to see how well you can refine these models that were not generated using Rosetta.

In this design puzzle we would like you to redesign the core. A few of the loops are locked and there are constraints to keep the backbone from moving too much, so focus on mutating the sidechains. If you are new to Foldit, make sure you have completed the Intro Design Puzzles 6-1 through 6-4.

We are reposting puzzle 350: CASP9 Refinement Puzzle 8A, because the native structure still has not been released. This time, however, we would like to see if these few constraints are helpful. This was the refinement target with 2 different starting models so again there will be multiple puzzles.

We are giving you this currently unsolved protein as an extended chain. Use all your skills to fold it up as best as you can. The secondary structure predictions are posted in the puzzle comments, but we have included them in the starting structure as well.
