This is a throwback puzzle to the early days of Foldit. This short sequence is a domain of a succinyltransferase enzyme in E. coli. We are revisiting old Foldit puzzles so we can see how useful the recent additions to the game have been. Players will NOT be able to load in any previous solutions for these puzzles.

This is a puzzle where you can redesign part of a player-designed protein. This is the subunit of a symmetric tetramer designed by jamiexq, karstenw, and markm457 for Puzzle 725. Baker lab simulations suggest that one of the beta strands is unlikely to form because the loops on either end prefer different conformations. Help us redesign these loops to find a solution that will fold up properly! You have one additional residue that you can insert in the protein, but most of the protein is frozen in this puzzle. This puzzle will only be active for three days!

We've analyzed the top solutions from the latest puzzle, in which you tried to fold an extended chain into an electron density cloud, and we want to see if you can keep improving on this structure. This puzzle includes the three most promising Foldit solutions (by Galaxie, meatexplosion, and karstenw) as starting structures, and the electron density cloud is a little more coarse. We're also providing you with a structure that was determined by another lab (PDB entry 3j40), although we're fairly certain this solution is inaccurate. You can load in your solutions from the previous puzzles (737, 740, 749, and 756) and can find more details in the puzzle comments.

This is a 45 residue symmetric pentamer design puzzle; there will be five symmetric chains to manage. Note that the score of a symmetric protein relies not only on how well-folded each chain is, but also on how well the chains interface together. See the blog post about this for more information. There are 2 filters enabled on this puzzle: the Residue IE Score monitors that all PHY, TYR, and TRP residues are scoring well, and the Core Exists filter checks that at least 30% of the residues are buried in the core of your design. The Baker Lab will run folding simulations on your solutions for this puzzle, and those that perform well will be synthesized in the lab. Remember, you can use the Upload for Scientists button for up to 3 designs that you want us to look at, even if they are not the best-scoring solutions.

The only aspect which is different in this quick puzzle is that the disulfide count filter is present, so you will have the chance to form disulfide bonds and get bonus points. This quick puzzle will only be up for a few days.
