This is a 91 residue symmetric dimer design puzzle; there will be two symmetric chains to manage. Note that the score of a symmetric protein relies not only on how well-folded each chain is, but also on how well the chains interface together. See the blog post about this for more information. There are 2 filters enabled on this puzzle: the Residue IE Score monitors that all PHY, TYR, and TRP residues are scoring well, and the Core Existence filter checks that at least 30% of the residues are buried in the core of your design. The Baker Lab will run folding simulations on your solutions for this puzzle, and those that perform well will be synthesized in the lab. Remember, you can use the Upload for Scientists button for up to 3 designs that you want us to look at, even if they are not the best-scoring solutions.

This is a throwback puzzle to the early days of Foldit. This protein is used by many viruses to insert their DNA into the human genome. We are revisiting old Foldit puzzles so we can see how useful the recent additions to the game have been. Players will NOT be able to load in any previous solutions for these puzzles.

This is Round 2 for Puzzle 748. You will be able to load in your manual saves from 748 and use them as a starting point here. This puzzle has been opened up to allow for sharing and the use of all scripts. We're also providing you with two templates in the alignment tool, and there will be one more round with server predictions. NOTE: If you did not manually save a solution in puzzle 748, you can go back to 748, manually save it, and the solution should appear in your manual saves for this puzzle.

We have trimmed the huge density cloud from Puzzle 746 by using your top-scoring solutions from Round 1. This will hopefully make this round a lot easier for everyone! You will be able to load in your manual saves from the previous round. NOTE: If you did not manually save a solution in puzzle 746, you can go back to 746, manually save it, and the solution should appear in your manual saves for this puzzle.

We are now giving you 10 server predictions for this unsolved protein from Puzzles 737 & 740. Resetting the puzzle will cycle through 10 different Quark Server predictions and they are available in the Alignment Tool so you can use partial threading between them. We are giving you a few extra days to work on this puzzle since there are 10 different models, so please try out each start as the topology that initially scores poorly may actually be the closest to the correct unknown structure. We are giving you another currently unsolved protein with no homologs as an extended chain. This round only GUI scripts are allowed and sharing has been disabled. After this puzzle expires, the puzzle will be re-posted and LUA scripts and sharing will be allowed. You'll be able to load in your solutions from this puzzle and use scripting and sharing. The secondary structure predictions are provided on the starting model, with more details posted in the puzzle comments.

This Electron Density puzzle is going to be as hard as it gets, which is why we are splitting it into 2 rounds. For this round, we have not trimmed the density. Again, the automated methods were unable to solve this structure using this huge density cloud. We will trim this density in Round 2 based on your top-scoring solutions for this round! Unfortunately there are no available templates to start with for this puzzle, so you'll have to use all your Electron Density folding skills to fit the density as well as you can! The secondary structure predictions have been loaded onto the starting model, with more details posted in the puzzle comments.
